Option-Return at the end of an expression prefixed with #_ is no longer triggering evaluation for me. If I use (comment ...) then it still works. Is this some new problem on my end, or an intentional change, or a regression, or... ?

It’s an unintended change. Please file an issue. I hope it still works if you have thee cursor inside the ignored form?
